Bioactivity Comments
IL-23 binds a heterodimeric receptor comprising an IL-12RP1 subunit (shared with the IL-12 receptor) and a unique receptor subunit, IL-23R. Receptor engagement activates JAK2 and Tyk2 to stimulate STAT1, -3, 4 and -5 signalling [5]. IL-23 plays an important role in bridging the innate and adaptive arms of the immune response [3].
The rationale for targeting the IL-23/IL-17/T helper type 17 (Th17) cell axis in the search for novel therapeutics for treating autoimmune inflammatory diseases is reviewed by Tang et al. (2012) [6], and further analysis of the role of IL-12 family cytokines in infection, inflammation and autoimmune disease is provided in [2].
