Comment: ABS-752 (also known as CT-01) is a small molecule GSPT1 and NEK7 molecular glue degrader [1]. It was designed to investigate the anti-tumour potential of reducing GSPT1 (GSPT1; P15170) protein levels. It is a prodrug that is converted by the monoamine oxidase VAP-1 (AOC3; which is overexpressed in diseased liver); the active molecule being ABT-002. ABT-002 induces formation of the degron/target/CRBN ternary complex, which promotes target protein degradation.
